چکیده

Research was conducted to determine levels of Cry1Ac in transgenic Bacillus thuringiensis Berliner (Bt) cotton leaves from four different commonly used bioassays. Significant differences in the amount of Cry1Ac were found among the different bioassay types. For bioassay arenas that prevented desiccation of leaf material, Cry1Ac levels remained stable for 10 d after tissue was excised from the plant. In other bioassay types, Cry1Ac levels increased in excised leaves over time because of desiccation that concentrated the transgenic protein. We conclude that careful descriptions of bioassay methodology are essential for laboratory studies documenting Cry1Ac levels and corresponding efficacy against lepidopteran pests.

PCR CONFIRMATION OF THE cry1Ac GENE IN TRANSGENIC BT (Bollgard) COTTON

Transgenic cotton (Gossypium hirsutum L.) with Bacillus thuringiensis Berliner δ-endotoxin genes has been widely adopted for suppression of lepidopteran pests. To account for inconsistency of the endotoxin gene expression, we developed a polymerase chain reaction (PCR) for checking the purity of transgenic cotton plants. A total of eight cry1Ac genes were aligned for the PCR primer design. A DN...

Production and characterization of Bacillus thuringiensis Cry1Ac-resistant cotton bollworm Helicoverpa zea (Boddie).

Laboratory-selected Bacillus thuringiensis-resistant colonies are important tools for elucidating B. thuringiensis resistance mechanisms. However, cotton bollworm, Helicoverpa zea, a target pest of transgenic corn and cotton expressing B. thuringiensis Cry1Ac (Bt corn and cotton), has proven difficult to select for stable resistance. Two populations of H. zea (AR and MR), resistant to the B. th...

Novel Pink Bollworm Resistance to the Bt Toxin Cry 1Ac: Effects on Mating, Oviposition, Larval Development and Survival

Bt cotton plants are genetically engineered to produce insecticidal toxins from the Bacillus thuringiensis (Bt) Berliner (Bacillales: Bacillaceae) bacterium and target key lepidopteran pests. In all previous strains of pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae) selected in the laboratory for resistance to insecticidal Cry1Ac toxin using an artificial diet cont...

Potential factors impacting season-long expression of Cry1Ac in 13 commercial varieties of Bollgard® cotton

Thirteen commercial varieties of transgenic Cry1Ac Bacillus thuringiensis Berliner (Bt) cotton were examined across two sites in 2000 for potential factors that impact endotoxin expression. In all cases, two varieties (NuCOTN 33B and DP 458B/RR, Delta & Pineland Co., Scott, MS) expressed more Cry1Ac than the other 11 varieties in various plant structures. These two varieties share the same pare...

Selective feeding of tobacco budworm and bollworm (Lepidoptera: Noctuidae) on meridic diet with different concentrations of Bacillus thuringiensis proteins.

Laboratory experiments were conducted to evaluate the behavior of bollworm, Helicoverpa zea (Boddie), and tobacco budworm, Heliothis virescens (F.), larvae on meridic diet with different concentrations of the Cry1Ac and Cry2Ab proteins from Bacillus thuringiensis subsp. kurstaki Berliner. The proteins used in these experiments are the ones in commercially available Bollgard and Bollgard II cott...
